Studs and Quick nuts

This sound like the title of a porn video. I know there have been a million posts on this. Anyway, I've searched the forum and read the previous posts, ad nauseam, and still don't completely get it. I might be a little slow and definitely mechanically impaired, so please bear with me. I definitely like the idea of being able to remove the door tops easily, but would like to understand the nuances of the stud/quick nut combo. Specifically, does this require any new holes? I believe it requires removing the glass to install (correct?), and if so, is there any difficulty or problems with water intrusion once the windows and/or door top are reset with the quick nuts in place? I don't want to create leak issues that currently aren't there (ie. if it ain't broke, don't fix it), but I think quick door top removal sounds really cool. Any feedback will really help and pics would be great.

Does that second "no" mean I don't have to remove the glass?

Bill Larson....you da man! It's all your fault that I'm ignoring my wife and kids to research studs and nuts.
