Just to be clear here, say I wanted to completely remove the oil cooler lines temporarily while waiting to get new ones, I could simply take the filter off, remove the plate, and refit oil filter (filled).

Correct?

What is the chance of loosing prime doing this? I would assume low since I'm not replacing the lines which are full of air.

There is a chance, I've lost prime while doing just that.

There is a chance, I was lucky and did not lose prime.
I did it because my line bursted and needed to run the truck while getting my act together to get the missing parts to install the new lines I had purchased long before that happened. I then ran the D90 without oil cooler/line for over a year (I only drive it "leisurely").

Make sure you blank the oil cooler radiator plugs correctly so nothing gets in there! Also, the oil filter will not be seated on a large flat surface like it is with the plate. Make sure you don't overtighten it.

Follow-up Post:

BTW, when I replaced my line, I filled the radiator + lines with oil (kept the same oil filter, topped it with oil also). It was messy, but I sure only wanted to take the minimum chance to lose prime. That would have increased the 30 minutes into a few hours (and just before leaving for MAR and a "few" more things to do..... That would not have been good).

Success in not loosing prime while changing oil and removing the sandwich plate. I filled the oil filter all the way to the top (letting all the air out) and the light went out almost instantly . I would assume if you were fitting new lines you would want to fill those with oil from the top when refitting to minimize air in the system, but I'm going without a cooler for now until I can get my hands on some new ones.
